a. Expected results

Marseilles :
[§ 1] The three sampling methods will be tested (random sampling, permanent sampling, and sampling using a positioning frame). This implies the following number of photo-quadrats expected :

1 transect? on the main site? x 3 sampling methods (permanent / random/random positioning frame) x 3 replicates x 20 photos = 180 photo-quadrats

b. Data analyses
Image processsing with the software PhotoQuad

[§ 2] Image processing will be carried out through the software PhotoQuad. (http://www.mar.aegean.gr/sonarlab/photoquad/index.php). This software of 2D image analysis is equipped with tools of spatial analysis (such as cover measuring), and species counts.

[§ 3] The identification of species, will be done manually at first. For this, it’s possible to start by applying, on each photograph, 100 stratified points. Then, each point is associated with a species or a category (rock, sediment, holes, etc.) from a library predefined and refillable as and when data processing with PhotoQuad (Figure 1).

[§ 4] When processing photographs, non-exploitable zones of the pictures should not be taken in account for the analysis of metrics : such as dark corners, unidentifiable species…
Photographs should be of sufficient quality to enable the identification of species : this level of quality is to determine during the beginning of the protocol implementation. It should represents the best quality/cost ratio (in terms of resolution, brightness, colour, …)

c. Statistical analysis

[§ 5] Statistical analyses will be performed using the R software. We will focus on taxa’s presence/absence, percent cover, taxa assemblages, and relations among between taxa’s abundances, and they will also be compared to environmental data.
